The Handkerchief is an item found in The Colonel's Bequest. It can be found starting in Act II, on the floor in the upstairs hallway.
The only purpose of the handkerchief is to be a clue for the murder, and can be examined with the monocle to determine that it has an "EP" embroidered on it in small letters. This would indicate it belongs to Ethel Prune.
